    Description

Sirolimus (Rapamycin, AY-22989) is a potent and specific mTOR inhibitor that binds to FKBP-12 to form a complex which inhibits mTORC1 activation. It inhibits mTOR activity in HEK293 cells with an IC₅₀ of approximately 0.1 nM and in mouse TSC1⁻/⁻ MEF cells with an IC₅₀ of 0.05 nM. Sirolimus is a 31‑membered triene macrolide antibiotic. It was first isolated from Streptomyces hygroscopicus obtained from a soil sample on Easter Island (Rapa Nui) and was initially identified as an antifungal agent. Sirolimus is widely used in biological research as an immunosuppressant to prevent organ transplant rejection, as an inducer of autophagy, and as a tool for chemically induced dimerization (CID) in studying protein‑protein interactions.
