    Description

Romidepsin (FK228, FR901228, NSC 630176) is a potent and selective class I histone deacetylase (HDAC) inhibitor with IC₅₀ values of 1.6 nM for HDAC1, 3.9 nM for HDAC2, and weaker activity against HDAC4 (25 nM) and HDAC6 (79 nM). It is the most potent HDAC inhibitor against Ewing sarcoma cell lines. Romidepsin is a bicyclic depsipeptide (cyclic peptide) isolated from Chromobacterium violaceum (and also from Burkholderia). In research and clinic, it is FDA‑approved for cutaneous and peripheral T‑cell lymphoma (Istodax®) and is widely studied as an epigenetic modulator in cancer and immunotherapy.
