It competitively inhibits ATP binding to HSP90 and downregulates NF‑κB signaling via IKKα/β at 1 μM in CLL cells. Alvespimycin is a hydroquinone ansamycin derivative of geldanamycin, prepared by demethylation and amination of geldanamycin. In preclinical research, it has shown activity against multiple myeloma and solid tumors, and is used to study the role of HSP90 in cancer and neuroprotection.
