    Description

Doxorubicin (Adriamycin, NSC 123127) is a topoisomerase II poison that intercalates DNA and stabilizes the Top2‑DNA cleavage complex, with activity dependent on Top2A expression. It inhibits DNA and RNA synthesis. Doxorubicin is an anthracycline antibiotic isolated from Streptomyces peucetius var. caesius. In research and clinic, it is one of the most widely used anticancer agents, employed as a chemotherapy drug for breast, ovarian, and many other cancers, and also as a positive control for DNA damage and apoptosis studies.

    Biological activity
Biological target: DNA Damage Response; Topoisomerase II (Top2A); Topoisomerase II (DNA intercalation)
In vitro activity: Intercalates DNA; stabilizes Top2‑DNA cleavage complex; Top2A expression correlates with sensitivity.
In vivo activity: Suppression of Top2A causes resistance; CD26/DPPIV enhances sensitivity in B‑cell lymphoma xenografts.
